Endocannabinoid turnover in GtoPdb v.2023.1

چکیده
The principle endocannabinoids are 2-acylglycerol esters, such as 2-arachidonoylglycerol (2-AG), and N-acylethanolamines, anandamide (N-arachidonoylethanolamine, AEA). glycerol esters ethanolamides synthesised hydrolysed by parallel, independent pathways. Mechanisms for release re-uptake of unclear, although potent selective inhibitors facilitated diffusion across cell membranes have been developed [29]. FABP5 (Q01469) has suggested to act a canonical intracellular endocannabinoid transporter in vivo [17]. For the generation 2-arachidonoylglycerol, key enzyme involved is diacylglycerol lipase (DAGL), whilst several routes synthesis described, best characterized which involves N-acylphosphatidylethanolamine-phospholipase D (NAPE-PLD, [75]). A transacylation forms N-acylphosphatidylethanolamines identified cytosolic enzyme, PLA2G4E (Q3MJ16) [66]. In vitro experiments indicate that also substrates oxidative metabolism via cyclooxygenase, lipoxygenase cytochrome P450 activities [5, 24, 77].

Nerve growth factor scales endocannabinoid signaling by regulating monoacylglycerol lipase turnover in developing cholinergic neurons.
Endocannabinoid, particularly 2-arachidonoyl glycerol (2-AG), signaling has recently emerged as a molecular determinant of neuronal migration and synapse formation during cortical development. However, the cell type specificity and molecular regulation of spatially and temporally confined morphogenic 2-AG signals remain unexplored. Here, we demonstrate that genetic and pharmacological manipulat...
متن کاملAstrocytes in endocannabinoid signalling.
Astrocytes are emerging as integral functional components of synapses, responding to synaptically released neurotransmitters and regulating synaptic transmission and plasticity. Thus, they functionally interact with neurons establishing tripartite synapses: a functional concept that refers to the existence of communication between astrocytes and neurons and its crucial role in synaptic function...

The signaling capacity of endogenous cannabinoids ("endocannabinoids") is tightly regulated by degradative enzymes. This Perspective highlights a research article in this issue (p. 996) in which the authors show that genetic disruption of monoacylglycerol lipase (MAGL), the principal degradative enzyme for the endocannabinoid 2-arachidonoylglycerol (2-AG), causes marked elevations in 2-AG level...
متن کاملEndocannabinoid signaling in the brain.
The primary psychoactive ingredient in cannabis, Delta9-tetrahydrocannabinol (Delta9-THC), affects the brain mainly by activating a specific receptor (CB1). CB1 is expressed at high levels in many brain regions, and several endogenous brain lipids have been identified as CB1 ligands. In contrast to classical neurotransmitters, endogenous cannabinoids can function as retrograde synaptic messenge...

The ECs (endocannabinoids) AEA (anandamide) and 2-AG (2-arachidonoylglycerol) and their lipid congeners OEA (N-oleoylethanolamide) and PEA (N-palmitoylethanolamide) are multifunctional lipophilic signalling molecules. The ECs, OEA and PEA have multiple physiological roles including involvement in learning and memory, neuroinflammation, oxidative stress, neuroprotection and neurogenesis. They ha...
